最近要使用PluriTest网页工具去计算细胞的Pluripotency，但是苦于这个网页工具的使用方法早在2011年已经发布，而网页工具已经有了较大的改动，缺少fastq.gz数据上传的详细protocol，在经过摸索之后，现将protocol呈现如下：
what's PluriTest?
PluriTest is a bioinformatic assay for pluripotency assessment. PluriTest.org provides an online platform for public to access and test the pluripotency for human PSCs of their interest
Protocol to proceed the assay
1. Open the website pluritest.org
2. Register and log in

3. Acceptable data format:
(1) Affymetrix HG-U219 (*.cel) files;
(2) FASTQ as gzipped (*.fastq.gz) for RNA-Seq, which is the format of our data；
(3) Illumina HT12v3 and HT12v4 (*.idat) files for the Illumina platform.
4. upload fastq.gz data
A. click “Get Started"

B. read the introduction and click the "Upload Files" button

C. choose the file type then submit

5. download the FTP Client FileZilla according to their recommendation
A. click the website, download and install the software as usual.
B. open the FileZilla Client and login
- Open FileZilla and enter the following in the Host field: ftpes://pluritest.org
- Enter your PluriTest login credentials in the Username and Password fields, unless you have a separate FTP password.
- Enter 21 in the Port field
- Click "Quickconnect"
- Locate the pending analysis FTP folder with name b7908caa-b359-422b-a4ee-207abcdd5429 (just an example) and double click it
- You can now drag and drop files to the remote folder pane on the right
- You can also use the left pane to select files to add to the upload queue and process them
- You can monitor the upload progress using the status pane at the bottom
- A maximum of 12 files are allowed per analysis. Any additional files will be ignored.
- Only *.fastq.gz files are accepted. All other files will be ignored.
- Please refer to the documentation for more info on using FileZilla Client

C. initiate the analysis
When you are finished uploading files, click here or check your Pending Analyses to initiate analysis (just for an example).
You must manually initiate the analysis after you are finished uploading your files
