摘要:
通过飞蝗基因组和转录组分析,作者发现了一个在雌性飞蝗成虫中高表达的基因Greglin。Greglin在雌性飞蝗脂肪体和卵巢中高表达,在卵黄发生期的表达量仅次于两个卵黄原蛋白(Vg)基因。而且,Greglin的表达受保幼激素的诱导。RNA干扰Greglin导致Vg蛋白的大量减少,显著抑制卵成熟、产卵量和卵孵化率。酶连免疫吸附实验表明,Greglin是通过抑制丝氨酸类蛋白酶活性而保护Vg不被水解,从而保障卵成熟所需的大量Vg。解析了丝氨酸蛋白酶类抑制剂Greglin在保护卵黄原蛋白白和卵形成中的重要作用。
